Despite recent advances in single cell sequencing, spatial distribution is still limited to broad categories such as cytoplasmic, nuclear or chromatin bound, and individual molecule resolution is not achievable. Single molecule RNA FISH (smFISH) technology is a highly complementary imaging method that provides this important additional information.
Dr. Susan Duncan, working in John Innes Centre, UK, is a distinctive specialist in developing smFISH in plants, she developed a new smFISH tech which uses pools of ~40 fluorescently labeled 20mer DNA probes to label individual RNA. Together these probes generate a ~300 nm diameter punctate spot of fluorescence that can be imaged and quantified at a subcellular level. Adaptation of smFISH for Arabidopsis root tissue has already enabled investigation into several aspects of plant gene regulation. Ongoing innovations to expand smFISH to a wider range of tissues and species will make it possible to explore even more aspects of plant RNA biology in the future.
